Fiber probe configurations simulation for depth-resolved skin fluorescence registration

Paper Abstract

Fiber-optic probes are widely used in biomedical research as convenient and compact devices for instrument-tissue interaction analysis. This paper presents a simulation of various configurations of fiber optic probes that implement selective registration of the spectral response from skin layers. The impact of the registration channel of the optical scheme on the skin fluorescence spectrum was analyzed upon excitation by laser radiation with a wavelength of 405 nm. A model of human skin has been developed, containing the main endogenous chromophores of the skin for a specific excitation wavelength. We compared the model skin fluorescence spectra with experimental spectra from real tissue. The propagation of excitation and fluorescence radiation in the system was simulated by the Monte Carlo method. It has been observed that it is necessary to take into account the optical design and the efficiency of collecting fiber probe in examining the spectral response from a multilayer biological tissue such as human skin. Because for fixed parameters of the tissue site under investigation, the recorded fluorescence spectrum at the detector varies depending on the sensitivity of the probe to individual layers of the biological tissue.
