Interferometric studies of a molecular machine

Paper Abstract

An adaptation of holographic interferometry to microscopic systems is described. The adaptation is suitable for studying motion even at the molecular level. The interferograms are fringeless, and they are presented in a form that discriminates against bulk translation, while mapping local displacements directly. The mapping is inherently insensitive to inhomogeneities in the illumination beam. Statistical interrogation of the maps is therefore unusually free of propagated error; and the effect of reconstruction noise, whose explicit calculation is given, is made transparent. The methods are illustrated by analyzing the motion of force-generating molecules of muscle. It is shown that their movements are correlated to a high degree.
