Paper Abstract

Flow cytometers based on elastic light scattering or in combination with some other optical methods are widely used in biological and medical investigations, though their use in practical medical laboratory diagnostics is less common. This seems to be due to the traditional application in such devices of the principle of hydrodynamic focusing, which leads to some deficiencies, namely: a) the complexity of the hydrodynamic focusing system; b) the necessity of a ballast pure liquids; c) the complexity and low reliability of the hydrodynamic system alignment with respect to the focused laser beam - a sight shift of the laser beam relative to the investigated liquid flow axis can fundamentally change the metrological possibilities of the device; d) the limitation upon the maximum size of cells to be measured (not more than the diameter of the investigated liquid flow). The aim of this paper is to study the possibility of avoiding these deficiencies by means of the laser probe-focused beam space scanning.
